ดัดแปรแป้งมันสำปะหลังโดยย่อยด้วยเอนไซม์แอลฟาอะไมเลสร่วมกับกลูโคอะไมเลส ที่ระดับความเข้มข้นรวมร้อยละ 0.139 (น้ำหนักเอนไซม์ต่อน้ำหนักแป้งแห้ง) โดยใช้ระยะเวลา 6, 12 และ 24 ชั่วโมง และดัดแปรแป้งมันสำปะหลังด้วยออกเทนนิลซัคซินิคแอนไฮไดร์ด 3 ระดับ คือ ร้อยละ 1, 2 และ 3 (ต่อน้ำหนักแป้งแห้ง) ศึกษาสมบัติทางกายภาพและเคมีฟิสิกส์ เช่น การเกิดเจลาติไนซ์เซชั่น, กำลังการพองตัวและร้อยละการละลาย, ลักษณะพื้นผิวของเม็ดแป้ง เป็นต้น นำสภาวะที่ให้สมบัติที่ดีที่สุดของการดัดแปรแป้งทั้งด้วยเอนไซม์และสารเคมีเพื่อทำการดัดแปรร่วมให้เป็นแป้งมันสำปะหลังออกเทนนิลซัคซิเนตที่ถูกย่อยด้วยเอนไซม์ โดยพบว่าการใช้สภาวะย่อยแป้งมันสำปะหลังเป็นระยะเวลา 24 ชั่วโมงและดัดแปรร่วมด้วยการใช้ออกเทนนิลซัคซินิคแอนไฮไดร์ดที่ระดับร้อยละ 3 (ต่อน้ำหนักแป้งแห้ง) เป็นสภาวะที่มีความเหมาะสมมากที่สุด โดยพบว่าแป้งมันสำปะหลังออกเทนนิลซัคซิเนตที่ถูกย่อยด้วยเอนไซม์มีค่าระดับการแทนที่สูงกว่าแป้งมันสำปะหลังออกเทนนิลซัคซิเนตที่ระดับร้อยละ 3 คือ มีค่าเท่ากับ 0.018±0.0001 และ0.015±0.0010 ตามลำดับ ลักษณะรูปร่างอนุภาคแป้งออกเทนนิลซัคซิเนตที่ถูกย่อยด้วยเอนไซม์มีการย่อยเกิดขึ้นทั้งบริเวณภายนอกพื้นผิวและภายในส่วนกลางของเม็ดแป้ง และเมื่อเปรียบเทียบขนาดของอนุภาคแป้งเฉลี่ยของแป้งออกเทนนิลซัคซิเนตที่ถูกย่อยด้วยเอนไซม์และแป้งมันสำหลัง พบว่าแป้งทั้งสองชนิดมีขนาดของอนุภาคเฉลี่ยไม่แตกต่างกันคือ มีค่าเฉลี่ยเท่ากับ 15.21 และ 15.17 ไมครอน ตามลำดับ อุณหภูมิการเกิดเจลาติไนเซชั่นของแป้งออกเทนนิลซัคซิเนตที่ถูกย่อยด้วยเอนไซม์มีค่าอุณหภูมิเริ่มต้นและอุณหภูมิที่จุดสูงสุดที่ใช้ในการเกิดเจลาติไนเซชั่นสูงกว่าแป้งมันสำปะหลังและแป้งออกเทนนิลซัคซิเนตอย่างมีนัยสำคัญทางสถิติที่ระดับความเชื่อมั่นร้อยละ 95 และพบว่าแป้งมันสำหลัง, แป้งที่ถูกย่อยด้วยเอนไซม์, แป้งออกเทนนิลซัคซิเนต และแป้งออกเทนนิลซัคซิเนตที่ถูกย่อยด้วยเอนไซม์มีค่าร้อยละการคืนตัวเท่ากับ 18.99, 23.65, 10.26 และ 7.62 ตามลำดับ เมื่อนำแป้งออกเทนนิลซัคซิเนตที่ถูกย่อยด้วยเอนไซม์มาผสมในน้ำสลัดไขมันต่ำ โดยลดปริมาณน้ำมันถั่วเหลืองและไข่แดงลงตั้งแต่ร้อยละ 10 ไปจนถึงร้อยละ 60 พบว่าสูตรที่ใช้แป้งดัดแปรที่ร้อยละ 2.8 (โดยน้ำหนัก) และลดปริมาณน้ำมันถั่วเหลืองและไข่แดงลงเหลือร้อยละ 20 (โดยน้ำหนัก) ได้รับการยอมรับทางประสาทสัมผัสและมีสมบัติทางกายภาพใกล้เคียงกับน้ำสลัดสูตรควบคุม ดังนั้นจึงมีความเป็นไปได้ที่จะนำแป้งออกเทนนิลซัคซิเนตที่ถูกย่อยด้วยเอนไซม์มาพัฒนาเป็นน้ำสลัดไขมันต่ำต่อไป
Modification of starch using enzymes and chemical were conducted. Starch was hydrolyzed by a mixture of α-amylase and glucoamylase at the total concentration of 0.139% (weight by weight) at 6, 12 and 24 hours. Then, esterification of octenyl succinic anhydride at 1, 2 and 3% (weight by weight) were applied. Physical and chemo-physic properties of starch, i.e. gelatinization, Swelling power, solubility and microscopic were determined. The results showed that the most proper condition for cassava starch modification in this study was enzymatic digestion for 24 hours followed by esterification of octenyl succinic anhydride at 3% (enzyme-hydrolysis octenyl succinylated cassava starch). Degree of substitution (DS) of enzyme-hydrolysis octenyl succinylated cassava starch was higher than octenyl succinylated cassava starch (p≤0.05), DS were 0.018±0.0001 and 0.015±0.0010, respectively. Enzyme-hydrolysis octenyl succinylated cassava starch granules with sponge-like and shells with the interior hydrolyzed was produced. Particle size distribution between enzyme-hydrolysis octenyl succinylated cassava starch and native cassava starch were not different (15.21 and 15.17 µm, respectively). The gelatinization temperature of enzyme-Hydrolysis octenyl succinylated cassava starch was significantly higher than native and octenyl succinylated cassava starch (p≤0.05). The retrogradation of native, enzyme-hydrolysis, octenyl succinylated and enzyme-hydrolysis octenyl succinylated cassava starch were 18.99, 23.65, 10.26 and 7.62 %, respectively. Applications of enzyme-hydrolysis octenyl succinylated cassava starch in 10 - 60 % fat-reduced salad dressing were tested. The resulted showed that using of enzyme-hydrolysis octenyl succinylated cassava starch at 2.8% by weight in 20% fat-reduced salad dressing had similar physical properties with control, and was accepted by sensory evaluation. Therefore, it indicated that enzyme-hydrolysis octenyl succinylated cassava starch could be potentially used as fat-reduced salad dressing.
